Offer Description

Among women, breast cancer (BC) is the most common cause of cancer, and the second leading cause of cancer death. The molecular classification of BC distinguishes luminal A and B expressing estrogen (ER) and/or progesterone receptors (PR), basal/triple negative breast cancer (TNBC), and human epidermal growth factor receptor 2 (HER2)-like tumors. This molecular taxonomy is clinically relevant with basal/TNBC patients presenting the poorest clinical outcome with no targeted therapies available when compared to other molecular subtypes. CD95 belongs to the tumor necrosis factor (TNF) receptor family and has mainly been viewed as a death receptor (Peter et al., 2015). However, recent data highlighted that this receptor can also induce non-apoptotic signaling pathways involved in physiological processes (Desbarats et al., 2003; Desbarats and Newell, 2000) or in the progression of auto-immune (O' Reilly et al., 2009; Poissonnier et al., 2016; Tauzin et al., 2011) and cancer disorders (Barnhart et al., 2004; Kleber et al., 2008). TNBCs maintain a very high amount of surface CD95 when compared to other breast cancers, and we recently found that CD95 loss in TNBC cells reprograms the immune landscape, by triggering a pro-inflammatory response unleashing the anti-tumor activity of natural killer (NK) cells. The CD95-driven molecular mechanism responsible for the recruitment and activation of NK cells in TNBC remains to be elucidated. This program proposes to address the molecular mechanism leading to NF-kB activation in CD95 deficient TNBC cells and hierarchize this signal using different molecular and biochemical assays.

Benefits

12 months contract (possibility to be renewed 12 monhs). Gross salary between 2600 and 3000 euros/months upon experience. Starting first semester 2022. Applicants are invited to send a CV (with list of publications, research experiences and referees)
